چکیده

Bis-quaternary ammonium compounds (bis-QACs) have the ability to cause a rapid and abundant leakage of the turbid materials from cells, and such a bacterioclastic ability leads to a potent bactericidal activity. In order to clarify the detailed mechanism of the bactericidal action of bis-QACs, the correlation between the bacterioclastic action of 4,4'-(1,6-hexamethylenedithio)bis(1-octylpyridinium bromide) (4DTBP-6,8) and the leakage of outer membrane pore protein E (OmpE) was investigated. Using the antiserum against a fusion protein consisting of GST and the OmpE protein of Escherichia coli encoded by the ompE gene, it was seen that the leakage of OmpE from E. coli cells was caused by treatment with low concentrations (much lower than the critical vesiculation concentration) of 4DTBP-6,8. Furthermore, it was confirmed that 4DTBP-6,8 caused an increase in the turbidity of the cell suspension of Klebsiella pneumoniae, Salmonella typhimurium and Serratia marcescences, and led to the leakage of several proteins which have a high percentage of homology with OmpE of E. coll. By immunoelectron microscopy investigation, it was revealed that the vesiculation from E. coli treated with 4DTBP-6,8 contains OmpE. In addition, the bacteriolytic action of 4DTBP-6,8 was investigated. The results suggested that the lysis of cells by bis-QACs was not an enzymatic action such as that by autolysin but a physical bacterioclastic action. Judging from these results, it is suggested that the leakage of OmpE is one of the major bacterioclastic actions of bis-QACs, and deals the bacterial cells a fatal blow.

Correlation between resistance of Pseudomonas aeruginosa to quaternary ammonium compounds and expression of outer membrane protein OprR.

The adaptation mechanism of Pseudomonas aeruginosa ATCC 10145 to quaternary ammonium compounds (QACs) was investigated. A P. aeruginosa strain with adapted resistance to QACs was developed by a standard broth dilution method. It was revealed that P. aeruginosa exhibited remarkable resistance to N-dodecylpyridinium iodide (P-12), whose structure is similar to that of a common disinfectant, cetyl...

AB INITIO QUANTUM CHEMICAL STUDIES IN CTAB

Quaternary ammonium compounds (QACS) are employed both as disinfectants for manual processing lines and surfaces in the food industry, and in human medicine area. QACS also cause cell death by protein denaturation . One of the QAC is cetyl trimethyl ammonium bromide (CTAB) that appears to rapture the cell membrane. The primary site of action of CTAB has been suggested to be the lipid components...

SDS-PAGE Analysis of the Outer Membrane Proteins of Uropathogenic Escherichia coli Isolated from Patients in Different Wards of Nemazee Hospital, Shiraz, Iran

Background: Outer membrane proteins (OMPs) constitute the main structure and about half of the cell wall of Gram-negative bacteria. The OMPs of Escherichia coli (E. coli) play an important role in its drug resistance. Previous studies have shown that the OMPs of E. coli enhance its pathogenic effects by helping the bacterium to evade the immune defense and promote its adsorption to host cells. ...

Effect of aminoacids on the fungicidal activity of quaternary ammonium salts

Amphipatic compounds exhibit an antimicrobial action both on bacteria and fungi. It is caused by a penetrative property of hydrophobic carbon chain of the compuound into a plasma membrane as well as by additional interaction of membrane elements and a hydrophilic amphipathic compound head. Bactericidal and fungicidal activity of this compound strongly depends on chemical environmental factors. ...
